| I. Background | |

CCR5（趨化因子受體 5 或趨化因子 C－C 亞族受體 5），也稱為 CD195，是一種 G蛋白 偶聯(lián)受體，屬于整合型膜蛋白的 β 趨化因子受體家族成員。CCR5 主要與 Gα i/o 和 Gα q/11 偶聯(lián)，在樹突狀細(xì)胞、T 細(xì)胞、巨噬細(xì)胞、嗜酸性粒細(xì)胞、小膠質(zhì)細(xì)胞和髓系抑制細(xì)胞上 表達(dá)，并由多種配體（CCL3、CCL3L1、CCL4、CCL5、CCL8、CCL11、CCL13 和 CCL16） 激活，以促進(jìn)趨化性和免疫細(xì)胞活化。
巨噬細(xì)胞和樹突狀細(xì)胞分泌 CCR5 依賴性促炎細(xì)胞因子可激活適應(yīng)性免疫反應(yīng)?；罨?的 CD4+ T 細(xì)胞分泌 CCR5 依賴性 IL-2 可誘導(dǎo)效應(yīng) T 細(xì)胞、記憶 T 細(xì)胞和調(diào)節(jié)性 T 細(xì)胞的 增殖和活化。 CCR5 是加速募集效應(yīng)和記憶 CD8+ T 細(xì)胞至感染部位所必需的。CCR5 是 HIV 進(jìn)入免疫細(xì)胞的輔助受體。CCR5 可與多種配體任意結(jié)合，從而啟動(dòng)包括引導(dǎo)遷移在 內(nèi)的細(xì)胞信號(hào)傳導(dǎo)。 |
